ABSTRACT

Objective To analyze the clinical characteristics of patients with human immunodeficiency virus (HIV) infection/acquired immunodeficiency syndrome (AIDS) and pulmonary tuberculosis, and investigate their survival and influencing factors of survival. Methods A total of 107 patients with HIV/AIDS and pulmonary tuberculosis were selected. The relationships of clinical symptoms, CT findings and CD4 cell count with positive laboratory tests were analyzed. Th survival of patients was investigated, and independent risk factors for death were analyzed. Results Most the 107 patients had symptoms such as cough, chest pain and fatigue. CT findings mainly included patchy shadow (75.70%), tree-in-bud sign (46.73%), nodular shadow (35.51%) and pulmonary hilar or mediastinal lymph node enlargement (86.92%). The proportion of lesions ≥ 3 pulmonary fields (47.66%) was higher. The positive rates of purified protein derivative (PPD), acid-fast bacilli and Mycobacterium tuberculosis were significantly higher in the CD4 cell count > 200/µL group than in the ≤200/µL group (P<0.05). There were statistically significant differences in body mass index (BMI), baseline CD4 cell count, multidrug resistant tuberculosis (MDR-TB) and standard anti-tuberculosis treatment between the survival group and the death group (P<0.05). Baseline CD4 cell count ≤200/µL, MDR-TB, and no standard anti-tuberculosis treatment were independent risk factors for death of patients with HIV/AIDS and pulmonary tuberculosis (P<0.05). Conclusion The clinical symptoms and imaging manifestations of patients with HIV/AIDS and pulmonary tuberculosis are complex and diverse, but characteristic. Baseline CD4 cell count ≤200/µL, MDR-TB and no standard anti-tuberculosis treatment are main risk factors for death of the patients.

ABSTRACT Objective: To determine the absolute number of serum T lymphocytes and cytokine levels and the characteristics of patients with active pulmonary tuberculosis and to assess their effect on the immune status of these patients and their diagnostic and predictive value for tuberculosis. Methods: We included 1,069 patients with active tuberculosis, 51 patients with latent tuberculosis infection, and 600 health individuals. Absolute serum T-lymphocyte counts and cytokine levels were quantified. Results: T lymphocytes were significantly reduced in patients with active tuberculosis when compared with healthy individuals. The immune function of patients gradually decreased with age and was stronger in female patients than in males. Th1 cells expressed higher levels of cytokines than did Th2 cells. Logistic regression analysis showed that reduced CD3+ T, CD8+ T, and NK cell counts, as well as reduced IL-4 and IFN-g expression, were independent influencing factors for active tuberculosis. ROC analysis showed that the sensitivity and specificity of absolute CD3+ T and CD8+ T lymphocyte counts and combined factors were significantly higher than were those of IL-4 and IFN-g for diagnosing active tuberculosis. Conclusions: Serum T-lymphocyte counts and cytokine levels can assess the immune status of tuberculosis patients; they are also useful biomarkers for predicting and diagnosing tuberculosis.


RESUMO Objetivo: Determinar o número absoluto de linfócitos T séricos e os níveis de citocinas séricas, bem como as características, de pacientes com tuberculose pulmonar ativa e avaliar o efeito desses no estado imunológico desses pacientes e seu valor diagnóstico e preditivo para tuberculose. Métodos: Foram incluídos 1.069 pacientes com tuberculose ativa, 51 pacientes com tuberculose latente e 600 indivíduos saudáveis. Foram realizadas a contagem absoluta de linfócitos T séricos e a quantificação de citocinas séricas. Resultados: Os linfócitos T estavam significativamente reduzidos nos pacientes com tuberculose ativa em comparação com os indivíduos saudáveis. A função imunológica dos pacientes diminuiu gradativamente com a idade e mostrou-se mais forte nas mulheres do que nos homens. As células Th1 expressaram maiores níveis de citocinas do que as células Th2. A análise de regressão logística mostrou que contagens reduzidas de células T CD3+, T CD8+ e NK e expressão reduzida de IL-4 e IFN-g foram fatores de influência independentes para tuberculose ativa. A análise ROC mostrou que a sensibilidade e especificidade dos valores absolutos de linfócitos T CD3+ e T CD8+ e de fatores combinados foram significativamente maiores do que as da IL-4 e do IFN-g para o diagnóstico da tuberculose ativa. Conclusões: A contagem de linfócitos T séricos e os níveis de citocinas séricas podem avaliar o estado imunológico de pacientes com tuberculose; também são biomarcadores úteis na predição e diagnóstico da tuberculose.

ABSTRACT

Objective:To study the relative molecular weight and distribution of polysaccharides in Polygonati Rhizoma (PR) before and after processing, as well as the effects of different polysaccharide fractions on immune function and inflammatory response of mouse peritoneal macrophages. Method:High-performance gel permeation chromatography (HPGPC) was used to determine the relative molecular weight and distribution of polysaccharides in PR (named SC) and polysaccharides in PR processed with wine (named JC), and polysaccharide fractions with different relative molecular weights were obtained by dialysis. Different polysaccharide fractions were applied to mouse peritoneal macrophages, which was normal or induced by lipopolysaccharide (LPS). Cell counting kit-8 (CCK-8) method was used to select the optimal administration concentration. Enzyme linked immunosorbent assay (ELISA) was used to determine the concentrations of tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>) and interleukin-1<italic>β</italic> (IL-1<italic>β</italic>) in the cell supernatant. The Griess method was used to detect the concentration of nitric oxide (NO). Result:SC and JC could be divided into four parts according to relative molecular weight and its distribution range, including part Ⅰ(14 800-2 273 kDa), part Ⅱ(2 148-296 kDa), part Ⅲ(12-1 kDa) and part Ⅳ(818-362 Da). Based on the differences of part Ⅰ and part Ⅲ after processing, the SC and JC were divided into two-part according to the weight-average relative molecular weight (<italic>M</italic><sub>W</sub>). For normal mouse peritoneal macrophages, JC could significantly promote the secretion of TNF-<italic>α</italic> (<italic>P</italic><0.01), while SC had no significant effect. Four polysaccharide fractions, named SD (SC fraction with <italic>M</italic><sub>W</sub>>50 kDa), JD (JC fraction with <italic>M</italic><sub>W</sub>>50 kDa), SX (SC fraction with <italic>M</italic><sub>W</sub><50 kDa) and JX (JC fraction with <italic>M</italic><sub>W</sub><50 kDa), also could<italic> </italic>significantly promote the secretion of TNF-<italic>α</italic> (<italic>P</italic><0.01), but only JX could significantly promote the secretion of NO (<italic>P</italic><0.05). In addition, the effect of JX group stimulated secretion of TNF-<italic>α</italic> was better than the JD group (<italic>P</italic><0.01). For the LPS-induced macrophage model, JC and SC group could<italic> </italic>significantly inhibit the secretion of TNF-<italic>α</italic> and IL-1<italic>β</italic> (<italic>P</italic><0.01), and the effect of JC was stronger. To compare different polysaccharide fractions, the impact of JX on inhibiting the secretion of TNF-<italic>α</italic> and IL-1<italic>β</italic> was<italic> </italic>significantly stronger than JD (<italic>P</italic><0.01), and SX inhibited the secretion of TNF-<italic>α</italic> was significantly stronger than SD (<italic>P</italic><0.01). Conclusion:The relative molecular weight and distribution of polysaccharides in PR before and after processing have changed. JC and SC improve the immune regulation mainly by inhibiting the inflammatory response, the fraction of <italic>M</italic><sub>W</sub><50 kDa is the main effective part, and the effect of PR polysaccharides in inhibiting the inflammatory is enhanced after processing with wine.

ABSTRACT

@#AIM: To analyze the consistency and accuracy of the axial position of the astigmatism correction intraocular lens(Toric IOL)measured by OPD scan Ⅲ(optical path difference analyzer)and the traditional slit lamp method.<p>METHODS: A prospective observational control study. A total of 118 patients with 156 eyes who underwent phacoemulsification combined with Toric IOL implantation in our hospital from July 2018 to October 2019 were selected. The residual astigmatism was followed up at 1wk, 1mo and 3mo after the operation, and the axial position of Toric IOL was measured with OPD scan Ⅲ under the small pupil(Axial intraocular astigmatism method)and after dilated pupils(Axial OPD method), while using traditional slit lamp method to measure Toric IOL axis position(Axial Slit method). Analyze the difference and consistency of the measurement results of the three methods, and calculate the Lens axis deviation(LAD)between the measurement results of the three methods and the target axis. <p>RESULTS: The residual astigmatism of the patients in this group was significantly lower than that before the operation at 1wk, 1mo and 3mo after operation(<i>P</i><0.05). The proportion of residual astigmatism ≤ 0.75D at 3mo after surgery was 73.7%. Three months after the operation, the axial position of the Toric IOL measured by the axial Slit method, the axial OPD method, and the axial intraocular astigmatism method were: 111.0°(10, 178)°, 113.5°(12, 180)°, and 113.0°(15, 178)°. Consistency analysis showed that the average value of the difference between the axial OPD method and the axial Slit method, the axial intraocular astigmatism method and the axial slit method, the axial OPD method and the axial intraocular astigmatism method at 3mo after the operation, they were -0.58°, -0.19°, 0.40°, which were all close to 0°, with high consistency. 95% <i>LoA</i> were(-7.01-5.84)°,(-12.44-12.07)°,(-10.69-11.49)°. At 3mo postoperatively, the proportions of patients with LAD ≤5° measured by axial Slit method, axial OPD method, and axial intraocular astigmatism were 82.0%, 80.1%, and 59.0%, respectively.<p>CONCLUSION: OPD scan Ⅲ can directly measure Toric IOL axial position after dilated pupils. It was an objective and accurate measurement method, which can replace the traditional slit lamp method to measure axial position and avoid subjective limitations. The Toric IOL axial position can also be measured by intraocular astigmatism under the small pupil, which has certain practical application value in ophthalmology clinical work.

ABSTRACT

ObjectiveSurgical site infection (SSI)is a type of common hospital-acquired wounds. The purpose of this study is to understand its clinical characteristics and prognosis in order to provide evidence for taking appropriate measures.Methods132 wound patients who met the diagnostic criteria of surgical site infection in the wound care center in recent 3 years were enrolled. The patients were treated locally with nanosilver dressing combined with red light and infrared ray. Individualized nutritional recipes and activity prescriptions were given, and the skin was showered every other day to heal or last 2 months.Observation indicators: demographics and wound characteristics; changes of area and depth after14 days intervention; the posi-tive rate of bacteria before and after the intervention, and healing rate and healing time of 2 months were analyzed and compared.ResultsA total of 132 cases of SSI(52.27% superficial and 47.73% deep ) was identified. The average age was 48.33±16.90 years. The average age and gender of the two groups were similar (P>0.05).Peri-wound skin is contaminated in all cases. The mean onset time of SSIs was 19.82±5.64 days, and the median onset time was 20 days. The onset time and duration of deep SSIs were longer than that of superficial SSIs (P0.05), and the depth of deep SSIs was deeper than that of superficial SSIs (P<0.05).The positive rate of bacterial culture inthe deep SSI group (79.31%) was higher than that of the superficial SSI group (59.46%) (P<0.05). After 14 days treatment, the area was smaller, the depth was lower (P<0.05), and the positive rate of bacteria was significantly decreased in the two groups (P<0.05). The healing rate ofsuperficial SSI group (84.06%) was higher than that of deep SSI group (66.67%) (P<0.05) in two months, and the healing time of superficial SSI group was significantly shortened (35.46<11.12 days vs 41.08<11.33 days, P<0.05). Logistic regression analysis showed that negative bacterial culture before intervention increased the healing index (OR=0.190). Long-term use of antibiotics did not promote healing (OR=0.343).ConclusionSuperficial and deep SSIs are common types of SSIs. Gender and age are similar, and skin contamination may play a role in the development of SSI,which needs attention. Local and systemic interventions can effectively improve wound healing. Negative bacterial culture and proper use of antibiotics can improve the healingprobability.

ABSTRACT

As the essential part of tratidional Chinese medicine(TCM), the research and development of classic formula have become a hot spot in TCM industry. However, with the change of the age, the species, medical part and origin of TCM have more or less changed. It is of great significance for the safety and effectiveness of the classical prescription to clarify the varieties and medicinal parts of TCM. In this paper, based on the discussion of the methods of textual research on the Chinese herbs, the species and medical parts, origin of Chinese herbs in a list of 100 famous classical formulas which promulgated by the state administration of TCM were analyzed. The textual research of Chinese herbs shows that most of the herbs involved in the classical formula have the problems of species, medical part, and origin. Therefore, it is of great significance for the selection of the species and medical parts, origin of the Chinese herbs in the research and development process of the classical formula.

ABSTRACT

Objective:To compare the total daily doses of 16 active components in big honeyed pills, concentrated pills and tablets of Fuzi Lizhongwan. Method:Three dosage forms of Fuzi Lizhongwan were prepared according to the process described in the literature. RRLC-QqQ-MS was employed to analyze the contents of 16 active ingredients with mobile phase of 0.1%formic acid aqueous solution-0.1%formic acid acetonitrile solution for gradient elution,the separation was performed on a Accucore RP-MS column(2.1 mm×100 mm, 2.6 μm) with a flow rate of 0.3 mL·min-1 and the column temperature at 30℃, the mass spectrometry condition was electrospray ion source, positive and negative ion switching mode for detection, multi-reaction monitoring mode(MRM) for scanning. The contents of 16 active ingredients were calculated, and the normalization arithmetic method was used for comparing the total daily doses of these active ingredients in three dosage forms of Fuzi Lizhongwan. Result:Processed products of Aconiti Lateralis Radix Praeparata were used as raw powder in preparation process of the three dosage forms, so there was no significant difference in the contents of six alkaloids in the three dosage forms, while the contents of other 10 active ingredients from Zingiberis Rhizoma, Codonopsis Radix, Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix et Rhizoma Praeparata cum Melle were significantly higher in big honeyed pills than those in concentrated pills or tablets(PConclusion:The total daily doses of 16 active ingredients in the three dosage forms of Fuzi Lizhongwan are significantly different caused by preparation process, prescription and dosage.

ABSTRACT

<p><b>Objective</b>To investigate the effect of Kangshuailing Gao (KG) on benign prostatic hyperplasia (BPH) in rats and its action mechanisms.</p><p><b>METHODS</b>Fifty BPH model rats were randomized into five groups of equal number, BPH model control, finasteride control, and high-, medium- and low-dose KG, to be treated intragastrically with distilled water, finasteride solution at 0.52 mg/kg, and KG solution at 4.16, 2.08 and 1.04 g/kg respectively once a day for 30 days consecutively. Another 10 normal healthy rats were taken as blank controls. The rats were weighed once a week during the treatment. The wet weight and index of the prostate were obtained after treatment, followed by measurement of the contents of serum estradiol (E2) and dihydrotestosterone (DHT), testosterone (T) and hypoxia-inducible factor-1α (HIF-1α) in the prostatic tissue, and observation of histomorphological changes in the prostate under the light microscope.</p><p><b>RESULTS</b>Compared with the BPH model control group, high- and medium-dose KG significantly reduced the prostate wet weight ([0.84 ± 0.08] vs [0.69 ± 0.04] and [0.71 ± 0.07] g, P < 0.01), the prostatic index ([0.28 ± 0.03]% vs [0.20 ± 0.02]% and [0.22 ± 0.03]%, P < 0.01), and the levels of T ([4.63 ± 1.25] vs [2.44 ± 0.47] and [2.91 ± 0.69] ng/L, P < 0.01) and DHT ([154.44 ± 20.25] vs [88.23 ± 13.63] and [90.52 ± 16.44] nmol/L, P < 0.01), but increased the level of E2 ([0.95 ± 0.24] vs [1.19 ± 0.14] and [1.20 ± 0.22] nmol/L, P < 0.01) in the serum. High-dose KG remarkably reduced the overexpression of HIF-1α in the prostate tissue of the BPH model rats (P < 0.01) and alleviated such BPH-related symptoms as epithelium thinning, intraglandular secretion reduction, and interstitial substance decrease.</p><p><b>CONCLUSIONS</b>Kangshuailing Gao acted effectively on BPH in the model rats by reducing the androgen level, balancing the estrogen/androgen ratio, and downregulating the expression of HIF-1α in the prostate tissue.</p>

@#AIM: To observe the clinical efficacy and safety of regional refractive multifocal intraocular lens SBL-3. <p>METHODS: Eighty-five senile cataract patients(130 eyes)who underwent phacoemulsification and intraocular lens implantation in our hospital were divided into two groups according to the difference of implanted intraocular lens: patients with regional refractive multifocal intraocular lens(MIOL)42 example(65 eyes)as an observation group, 43 patients(65 eyes)of a single-focus aspheric intraocular lens were used as a control group. Three months after operation, the indicators were: uncorrected distance visual acuity(UCDVA), uncorrected intermediate visual acuity(UCIVA), uncorrected near visual acuity(UCNVA), and spherical equivalent power(SE). The naked eye defocus curve was measured and the patient's subjective visual quality was investigated by questionnaire, including the rate glasses removal, visual interference symptoms and satisfaction. <p>RESULTS: At 3mo after operation, UCDVA was significantly improved in both groups compared with preoperative, and the difference was statistically significant(<i>P</i><0.01). However, there was no significant difference between the two groups(<i>P</i>>0.05), while UCIVA and UCNVA in the observation group were significantly better than the control group, the difference was statistically significant(<i>P</i><0.01). At 3mo after operation, the difference of computerized optometry spherical equivalent power(SE)of the two groups was statistically significant(<i>P</i><0.01). At 3mo after operation, the naked eye defocus curve of the observation group was stable in the range of +0.50 to -3.00D, the LogMAR visual acuity was below 0.301, and the curve began to rise slowly after -3.50D; the control group showed trough between ±0.5D, both ends are rising rapidly. At 3mo postoperatively, the proportion of patients with visual interference symptoms in the observation group(4/42)was not significantly different from that in the control group(2/43)(<i>P</i>=0.433). The glasses removal rate of the observation group was 97.6%(41/42), control group was 18.6%(8/43), the difference was statistically significant(<i>χ</i><sup>2</sup>=4.318, <i>P</i>=0.038). In the observation group, the single eye distance satisfaction was 92.3%, and the control group was 93.8%. The difference was not statistically significant(<i>P</i>=1.000). The observation group had a single eye near fullness of 98.5%(64/65)and the control group was 16.9%(11/65), the difference was statistically significant(<i>χ</i><sup>2</sup>=88.526, <i>P</i><0.01). <p>CONCLUSION: The regional refraction MIOL can provide good near-intermediate-distance and full-distance uncorrected visual acuity. It can meet the needs of the patients at daily, distance-middle distance-near use. Postoperative visual interference symptoms were fewer and patient satisfaction was higher.

Objective Few studies have paid attention to time-zero renal biopsy in living kidney transplantation so far. This article aimed to investigate the risk factors of latent pathologic changes in living donors by time-zero renal biopsy (TO-RBx) and the predictive value in the allograft function of recipients early after living kidney transplantation.Methods We retrospectively analysed the clinical data of 89 renal transplant recipients and living donors who received TO-RBx at Nanjing General Hospital from January 2008 to December 2016. According to the 2007 Banff criteria, the common pathologic changes in living donors such as latent glomeruloscerosis (GS), tubular atrophy (CT), interstitial fibrosis (CI), arteriolar hyaline thickening (AH) and vascular fibrous intimal thickening (CV) were scored. To analyze the influencing factors for different pathological changes and evaluate its predictive value in the allograft function of recipients in 1, 3, 6 months after living renal transplantation.Results Of all the TO-RBx specimens, 23 cases (25.84%) with GS (21 were mild change, 1 was moderate change and 1 was severe change), 33 cases (37.08%) with CT/CI changes (30 were mild change and 3 were moderate change) and 37 cases (41.57%) with AH/CV changes (36 were mild change and 1 was moderate change). GS was related to the donor age (P=0.042); CT/CI changes were related to donor age, gender and systolic pressure (P=0.019;0.006;0.01); arterial changes were related to donor gender and blood triglyceride level (P=0.029;0.049). Within 3 and 6 months after living donor renal transplantation, the eGFR of renal transplant recipients with GS lesions \[(65.96±17.17), (69.52±19.1)mL/min·1.73m2\] were significantly lower than the groups without lesions \[(76.91±18.98), (79.52±18.91)mL/min·1.73m2\] (P<0.05).Conclusion Time-zero renal biopsy has significance in terms of predicting the allograft function in 6 months after transplantation. It can guide the formulation and adjustment of postoperative immunosuppressive regimens for recipients. Besides, it can also detect the latent pathologic changes in living donors and is one of the important evidence for establishing a personalized follow-up plan for donors after surgery. This method is practical in clinical.

ABSTRACT

Placentation,which is critical for maternal-fetal exchange of nutrients and gases,is a complicated process comprising stepwise vasculogenesis and angiogenesis.Hypoxia caused by impairedtrophoblast invasion may cause various angiogenic abnormalities in human placenta.The Notchl signaling pathway plays an important role in the regulation of angiogenesis.The angiogenesis of human umbilical vein endothelial cells (HUVECs) under normal/hypoxic conditions and the mRNA/protein level of Notchl/Dell4/Jaggedl were investigated in this study.The effects of DAPT/JAG-1 on the migration of HUVECs were also assessed by cell wound healing assay,so as to discover the possible role of notchl signaling pathway in the angiogenesis of human placenta.The results showed that angiogenic ability of HUVECs was seriously reduced under hypoxic conditions.The mRNA and protein levels of Notchl/Dell4/Jaggedl were decreased in the hypoxic group compared to the control one.In addition,the migration capability of HUVECs was significantly obstructed when treated with DAPT and under hopoxic condition,but promoted when treated with JAG-1.The above results demonstrate that hypoxia downregulates the angiogenesis in human placenta via Notch 1 signaling pathway.

ABSTRACT

Background: Teratoma at the esophagogastric junction is extremely rare. Case Report: A 26-day-old male neonate who presented with vomiting and melena. Observation: Investigations revealed a mass at the esophagogastric junction, which was excised and confirmed to be a teratoma. Outcome: A fistula at the lower end of the esophagus and an esophageal hiatal hernia were observed as complications. Message: A careful surgical approach is warranted for a teratoma in esophagogastric junction, to avoid postoperative complications.

ABSTRACT

A male patient, 55 years old, was found from a container yard 65 h later following a chemical warehouse explosion in Tianjin, China on August 12, 2015. He was about 50 m away from the explosion center. He was subjected to compound multiple trauma, multi-viscera function damage, multiple fractures, hemothorax, traumatic wet lung, respiratory failure I, hypovolemic shock and impaired liver and kidney functions. After a series of successful treatments, he was rescued and recovered well.

ABSTRACT

The relationship between T cell immunoglobulin domain and mucin domain protein 3 (Tim-3)/Galectin (Gal)-9 pathway and recurrent spontaneous abortion (RSA) was studied. Thirty-one pregnant women with RSA and 27 normal early gravidas were investigated to detect the levels of Tim-3 and Gal-9 in villi and deciduas by Western blotting. Meanwhile, the concentration of interleukin (IL)-4 and IL-12 in peripheral blood plasma was determined by ELISA in 25 healthy fertile non-pregnant controls, the normal early gravidas and pregnant women with RSA mentioned above, respectively. It was found that the relative expression levels of Tim-3 and Gal-9 in villi and deciduas were significantly increased in pregnant women with RSA as compared with those in the normal early gravidas. The concentration of IL-4 in peripheral blood plasma of pregnant women with RSA was lower than that of the normal early gravidas (P<0.05) and healthy fertile non-pregnant controls (P<0.05), but that of IL-2 in pregnant women with RSA was significantly higher than that of the normal early gravidas (P<0.05) and healthy fertile non-pregnant controls (P<0.05). It was suggested that the overexpression of Tim-3/Gal-9 pathway may be related to the pathogenesis of RSA.

ABSTRACT

Objective To find out the status of drinking-water-borne endemic fluorosis and the effect of preventive measures in Hebei Province,so as to provide a basis to prevent and cure fluorosis.Methods One affected county (city,district) with drinking-water-borne endemic fluorosis was sampled in every city and 10 water improvement projects were investigated in that county.Three villages were taken out in every county.The operating state of the projects,the water fluoride content,and the dental fluorosis of children aged 8-12 and skeletal fluorosis of adults were investigated in these villages.Results A total of 93 projects were investigated,among which 94.6%(88/93) were operating properly and the qualified rate(fluoride content ≤ 1.2 mg/L) of the projects was 54.84%(51/93).Among the 9 large projects,the water fluoride content of 7 projects exceeded 1.2 mg/L.A total of 23 villages with water improvement projects were investigated and only 43.4% (10/23) of them water fluoride content were lower than 1.2 mg/L.The dental fluorosis rate,the dental fluorosis index and the rate of skeletal fluorosis were 33.2% (532/1601),0.74 and 5.7% (66/1155),respectively.In the 7 villages without water improvement projects,2 of them exceeded 2.0 mg/L,but lower than and equal to 4.0 mg/L of water fluorosis and 5 of them were normal(< 1.2 mg/L).The dental fluorosis rate,the dental fluorosis index and the rate of skeletal fluorosis were 33.4% (111/332),0.72 and 2.9% (10/350),respectively.Conclutions The qualified rate of water improvement projects of fluoride content ≤ 1.2 mg/L is low and the endemic fluorosis is still comparatively serious in Hebei Province.The progress of water improvement projects in the areas with endemic fluorosis should be accelerated and the quality of water improvement projects should be increased.

ABSTRACT

This study aimed to identify biochemical predictors of adverse perinatal outcomes in intrahepatic cholestasis of pregnancy (ICP). A total of 106 ICP cases were analyzed retrospectively by the combination of receiver operating characteristic curve and binary logistic regression analysis. "Adverse perinatal outcomes" included spontaneous preterm labor, meconium-staining of amniotic fluid, stillbirth and Apgar score ≤7 at 1 or 5 min. Total bile acid (TBA) [AUC=0.658, 95%CI (0.536, 0.781), P=0.031] was a valuable predictor for adverse perinatal outcomes. The critical value of TBA above which adverse perinatal outcomes were observed was 40.15 μmol/L (Youden's index=0.3). Binary multivariate logistic regression analysis revealed that the risk of adverse perinatal outcomes increased when TBA ≥40.15 μmol/L [OR=3.792, 95%CI (1.226, 11.727), P=0.021]. It is concluded that the risk of adverse perinatal outcomes in ICP increases when maternal TBA ≥40.15 μmol/L.

ABSTRACT

Human papillomavirus (HPV)-induced cervical cancer is the second most common cancer among women worldwide. Despite the encouraging development of the preventive vaccine for HPV, a vaccine for both prevention and therapy or pre-cancerous lesions remains in high priority. Thus far, most of the HPV therapeutic vaccines are focused on HPV E6 and E7 oncogene. However these vaccines could not completely eradicate the lesions. Recently, HPV E5, which is considered as an oncogene, is getting more and more attention. In this study, we predicted the epitopes of HPV16 E5 by bioinformatics as candidate peptide, then, evaluated the efficacy and chose an effective one to do the further test. To evaluate the effect of vaccine, rTC-1 (TC-1 cells infected by rAAV-HPV16E5) served as cell tumor model and rTC-1 loading mice as an ectopic tumor model. We prepared vaccine by muscle injection. The vaccine effects were determined by evaluating the function of tumor-specific T cells by cell proliferation assay and ELISPOT, calculating the tumor volume in mice and estimating the survival time of mice. Our in vitro and in vivo studies revealed that injection of E5 peptide+CpG resulted in strong cell-mediated immunity (CMI) and protected mice from tumor growth, meanwhile, prolonged the survival time after tumor cell loading. This study provides new insights into HPV16 E5 as a possible target on the therapeutic strategies about cervical cancer.
